【摘
要】目的 研究胰腺癌肿瘤干细胞体外增殖能力及其对化疗药吉西他滨耐受性。方法
以CD44+CD24+ESA+为表面标记,用流式细胞仪,在人胰腺癌Panc-1细胞系中分选肿瘤干细胞,接种在96孔板中,用CCK8法检测其体外增殖能力,描制生长曲线,用吉西他滨分别干预普通肿瘤细胞和肿瘤干细胞,比较分析化疗药刺激对两种细胞生长的影响,并计算IC50值。结果 CD44+CD24+ESA+阳性细胞生长速度明显慢于Panc-1细胞,呈干细胞球样缓慢生长,其对吉西他滨抑制试验不如Panc-1细胞敏感。结论 .肿瘤干细胞在体外培养中表现缓慢生长及为耐受化疗药物的干细胞特性。南通大学附属医院甲乳外科陈锦鹏

【Abstract】 Objective
To study the proliferation and drug -resistence ability of pancreatic
cancer stem cell in vitro. Methods Isolate pancreatic cancer stem cells of
CD44+CD24+ESA+ phenotypes by a FACS Aria II in human cell line Panc-1. To investigate proliferation ability of
cancer stem cell, we draw a growth curve by ways of CCK8 staining. To detect
the ability of drug-resistence of cancer stem cell, we stimulated both cells by
gemcitabine and study the difference of their IC50.Results Panc-1 Cell and CD44+CD24+ESA+ cells were restrained
by gemcitabine. In 48th hour IC50 of Panc-1 cell is 982μg/ml, IC50 of CD44+CD24+ESA+ is 1981μg/ml. In 72th hour IC50 of Panc-1 cell is 670μg/ml, IC50 of CD44+CD24+ESA+ cell is 1484μg/ml Conclusions Cancer stem cell perform a stem
cell-like characteristic of declined proliferation and highly drug-resistence.
